Mutations and blood clots: how point mutations in clotting factor genes conspire to increase the risk of thrombosis

چکیده

After trauma, the formation of a thrombus is essential to stem bleeding. A cascade of proenzymes, enzymes, and cofactors interact with damaged vessel endothelium to converge on a common pathway with the formation of a fibrin clot. The clot acts as a mechanical plug to prevent bleeding and is vital for normal vascular function. Disturbance of this pathway can be deadly; too little clotting results in bleeding disorders such as hemophilia, whereas excessive clotting produces blood clots that can block the lungs.

Factor V Leiden, MTHFR C677T and Prothrombin Gene Mutation G20210A in Iranian Patients with Venous Thrombosis

Background: Factor V Leiden, Prothrombin gene (G20210A) and MTHFR (C677T) polymorphism are the main biomarkers for evaluation of tendency for venous thromboembolism. We aimed to investigate the frequency of mutations in factor V Leiden, Prothrombin G20210A and MTHFR C677T and identify the genetic status for these mutations in patients with venous thrombosis. Methods: This study was carried out...

Nonsense-mediated mRNA decay among coagulation factor genes

Objective(s): Haemostasis prevents blood loss following vascular injury. It depends on the unique concert of events involving platelets and specific blood proteins, known as coagulation factors. The clotting system requires precise regulation and coordinated reactions to maintain the integrity of the vasculature. Clotting insufficiency mostly occurs due to genetically inherited coagulation fact...

Association of mtDNA mutation with Autism in Iranian patients

The autism spectrum disorders (ASD) are amongst the most heritable complex disorders. Although there have been many efforts to locate the genes associated with ASD risk, many has been remained to be disclosed about the genetics of ASD. Scrutiny's have only disclosed a small number of de novo and inherited variants significantly associated with susceptibility to ASD. These only comprise a small ...

Screening for Causative Mutations of Major Prolificacy Genes in Iranian Fat-Tailed Sheep

Objective The presence of different missense mutations in sheep breeds have shown that the bone morphogenetic protein receptor 1B (BMPR1B), bone morphogenetic protein 15 (BMP15) and growth differentiation factor 9 (GDF9) genes play a vital role in ovulation rate and prolificacy in ewes. Therefore, the present study investigates BMPR1B, BMP15 and GDF9 genes mutations in prolific ewes of Iranian ...

Inherited Genetic Markers for Thrombophilia in Northeastern Iran (a Clinical-Based Report)

Background: Thrombophilia is a main predisposition to thrombosis due to a procoagulant state. Several point mutations play key roles in blood-clotting disorders, which are grouped under the term thrombophilia. These thrombophilic mutations are methylenetetrahydrofolate reductase (MTHFR, C677T, and A1298C), factor V Leiden (G1691A), prothrombin gene mutation (factor II, G20210A), and plasminogen...
